Overview

Erfonrilimab (KN046) is an investigational bispecific monoclonal antibody developed by Jiangsu Alphamab Biopharmaceuticals. It is designed to simultaneously target and block two key immune checkpoint pathways: Programmed Death-Ligand 1 (PD-L1) and Cytotoxic T-Lymphocyte-Associated Protein 4 (CTLA-4). By dual-targeting these receptors, erfonrilimab aims to restore and enhance the anti-tumor activity of T cells while potentially offering a more favorable safety profile compared to combinations of separate PD-(L)1 and CTLA-4 inhibitors. It is currently being evaluated in Phase II and Phase III clinical trials for various solid tumors, including non-small cell lung cancer (NSCLC), esophageal squamous cell carcinoma, and, in combination with the HER2-targeted bispecific antibody KN026, for HER2-positive solid tumors such as gastric and breast cancers.
