Eribulin is a fully synthetic macrocyclic ketone analog of the marine natural product halichondrin B. It is an antineoplastic drug classified as a mitotic inhibitor. Eribulin is primarily used in the treatment of metastatic or locally advanced breast cancer and unresectable or metastatic liposarcoma, typically after failure of at least two prior chemotherapy regimens. Its mechanism of action involves binding to microtubule plus ends, inhibiting microtubule dynamics required for cell division, leading to irreversible mitotic blockade and apoptosis of cancer cells. Beyond its cytotoxic effects, eribulin also induces non-mitotic changes such as vascular remodeling, reversal of epithelial-mesenchymal transition (EMT), and reduced metastatic potential. The drug is administered intravenously.
