Eribulin + rebastinib is an investigational combination therapy being studied primarily for the treatment of HER2-negative metastatic breast cancer. Eribulin is a non-taxane microtubule inhibitor that disrupts microtubule dynamics and induces apoptosis in cancer cells. Rebastinib is a potent small molecule inhibitor of TIE2 kinase and ABL1 kinase; it blocks TIE2-mediated tumor microenvironment of metastasis (TMEM) doorway function, thereby inhibiting tumor cell dissemination and metastasis. The combination aims to leverage the cytotoxic effects of eribulin with the anti-metastatic properties of rebastinib by targeting both tumor cells and their supportive microenvironment[1][7][9]. Clinical studies have shown pharmacodynamic evidence for TIE2 inhibition with this regimen, as well as reductions in circulating tumor cells[1].
