Overview

This is a triple combination therapy consisting of eribulin mesylate (a non-taxane microtubule dynamics inhibitor) with dual HER2-targeted therapy (pertuzumab and trastuzumab). Eribulin mesylate inhibits tubulin polymerization, suppressing microtubule functions and inducing cell-cycle arrest at the G2/M phase, ultimately leading to apoptosis. Pertuzumab and trastuzumab are monoclonal antibodies that target different domains of the HER2 receptor, providing complementary mechanisms of HER2 blockade. This combination has been studied as an alternative to taxane-based regimens for HER2-positive breast cancer, particularly for patients who cannot tolerate taxanes or have developed resistance to them.
