This combination therapy consists of eribulin mesylate, pertuzumab, and trastuzumab. Eribulin mesylate is a non-taxane microtubule dynamics inhibitor that binds to high-affinity sites on the growing positive ends of microtubules, leading to irreversible mitotic block and apoptosis. Pertuzumab and trastuzumab are recombinant humanized monoclonal antibodies that target the human epidermal growth factor receptor 2 (HER2). Trastuzumab binds to domain IV of HER2, inhibiting ligand-independent signaling and mediating antibody-dependent cellular cytotoxicity (ADCC). Pertuzumab binds to domain II, preventing HER2 from dimerizing with other HER family members (HER1, HER3, HER4). This triplet combination is being investigated by Sun Yat-Sen Memorial Hospital as a neoadjuvant therapy for patients with HER2-positive early or locally advanced breast cancer to improve pathological complete response (pCR) rates.
