Overview

Erlotinib + dalotuzumab is a combination therapy investigated primarily for the treatment of non-small cell lung cancer (NSCLC) and other solid tumors. Erlotinib is a small molecule tyrosine kinase inhibitor that targets the epidermal growth factor receptor (EGFR), blocking its kinase activity and thereby inhibiting downstream signaling pathways involved in tumor cell proliferation and survival. Dalotuzumab (also known as MK-0646) is a humanized monoclonal antibody that targets the insulin-like growth factor 1 receptor (IGF1R), preventing ligand-induced activation, promoting receptor internalization and degradation, and inhibiting downstream PI3K/Akt and Ras/Raf/MEK/Erk signaling pathways associated with cell survival, proliferation, and resistance to apoptosis. The rationale for combining these agents stems from evidence of crosstalk between EGFR and IGF1R pathways in cancer cells; dual inhibition may enhance antitumor efficacy compared to targeting either pathway alone[2][4][6][8].
