Erlotinib + onartuzumab is a combination therapy consisting of erlotinib, an epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor, and onartuzumab, a monoclonal antibody targeting the hepatocyte growth factor receptor (MET). This combination was investigated primarily for the treatment of non-small-cell lung cancer (NSCLC), particularly in patients with MET-positive tumors. Erlotinib inhibits EGFR signaling to block tumor cell proliferation and survival, while onartuzumab blocks MET signaling implicated in resistance to EGFR inhibitors and poor prognosis. The rationale for combining these agents was to overcome resistance mechanisms mediated by MET activation. Clinical trials showed mixed results; phase II data suggested benefit in MET-positive NSCLC patients, but subsequent phase III studies failed to demonstrate improved outcomes compared to erlotinib alone[1][2][5][6].
