Ertumaxomab is a trifunctional bispecific monoclonal antibody designed to target both HER2/neu (human epidermal growth factor receptor 2) on tumor cells and CD3 on T cells. By binding simultaneously to these two antigens, ertumaxomab forms a tri-cell complex between tumor cells, T lymphocytes, and accessory immune effector cells such as macrophages, dendritic cells, and natural killer (NK) cells. This interaction redirects the immune system to attack HER2-positive cancer cells by activating T lymphocytes and accessory immune effector mechanisms including antibody-dependent cell cytotoxicity (ADCC), apoptosis induction, macrophage stimulation, NK cell activation, and overall immunostimulation. Ertumaxomab was primarily developed for the treatment of metastatic breast cancer in patients who have progressed after trastuzumab therapy[1][4][5][7][9].
