ERY974 is a humanized IgG4 bispecific monoclonal antibody developed as a cancer immunotherapy. It simultaneously binds to glypican 3 (GPC3), an oncofetal protein overexpressed in several tumor types, and CD3, a component of the T-cell receptor complex on cytotoxic T lymphocytes. By crosslinking GPC3-expressing tumor cells with CD3-positive T cells, ERY974 redirects and activates T cells to induce potent cytotoxicity against GPC3-positive cancer cells. This mechanism enables targeted lysis of tumor cells while sparing normal tissues where GPC3 expression is minimal. Preclinical studies demonstrated antitumor efficacy in various models, including those resistant to other immunotherapies. Clinical development focused on advanced solid tumors such as hepatocellular carcinoma, lung cancer, gastric cancer, esophageal cancer, and head and neck cancers[1][5][6][7].
