Eryaspase is a biologic drug consisting of the enzyme L-asparaginase encapsulated within donor-derived red blood cells (erythrocytes). Its mechanism of action involves the hydrolysis of plasma asparagine to aspartic acid and ammonia by the encapsulated L-asparaginase. This depletes circulating asparagine, an amino acid essential for protein synthesis in certain cancer cells that lack sufficient asparagine synthetase. By reducing plasma asparagine levels, eryaspase inhibits protein and nucleotide synthesis in tumor cells, leading to apoptotic cell death. Encapsulation within red blood cells prolongs enzyme activity and reduces toxicity compared to free L-asparaginases. Eryaspase is primarily developed for patients with acute lymphoblastic leukemia (ALL) who are hypersensitive to pegylated *E. coli*–derived asparaginases and has also been investigated for advanced or metastatic pancreatic ductal adenocarcinoma (PDAC) in combination with chemotherapy[1][3][4][6][7].
