**Erymethionase** is an experimental enzyme therapy consisting of methionine gamma-lyase (MGL), a bacterial enzyme that catalyzes the degradation of L-methionine into alpha-keto-gamma-methylthiobutyrate, ammonia, and methanethiol, encapsulated within autologous red blood cells (RBCs) to prolong circulation and reduce immunogenicity. Developed by ERYTECH Pharma using their ERYCAPS platform, it depletes systemic L-methionine levels, exploiting the methionine auxotrophy of certain tumors to induce tumor starvation while sparing normal cells. Preclinical studies demonstrate enhanced efficacy in combination with immune checkpoint inhibitors like anti-PD-1 by reducing adenosine in the tumor microenvironment and hypermethylation-mediated immune escape, with applications in triple-negative breast cancer (TNBC), glioblastoma, and homocystinuria.[3][6][9][18]
