ES005 is a humanized monoclonal antibody that targets Signal Regulatory Protein Alpha (SIRPα), an inhibitory receptor predominantly expressed on myeloid cells such as macrophages, monocytes, and dendritic cells. Developed by Elpiscience, ES005 is designed to block the interaction between SIRPα and CD47, a ligand frequently overexpressed on the surface of cancer cells as a "don't eat me" signal. By disrupting this checkpoint, ES005 relieves the inhibition of macrophage-mediated phagocytosis, thereby promoting the clearance of tumor cells. A key advantage of targeting SIRPα over CD47 is the potential for a superior safety profile; since SIRPα expression is largely restricted to myeloid cells and absent on erythrocytes, ES005 avoids the significant hematologic toxicities, such as anemia and thrombocytopenia, often observed with anti-CD47 therapies. ES005 is currently being evaluated in clinical trials for the treatment of advanced solid tumors, both as a monotherapy and in combination with other agents.
