ES014 is a first-in-class bispecific antibody fusion protein that targets both the CD39 and transforming growth factor-beta (TGF-β) pathways. It consists of a monoclonal antibody directed against CD39 (ectonucleoside triphosphate diphosphohydrolase 1) fused to the TGF-β receptor type II ectodomain. The drug is designed to reverse immunosuppression in the tumor microenvironment by inhibiting adenosine generation via CD39 blockade and neutralizing TGF-β signaling. This dual mechanism aims to convert immunologically "cold" tumors into "hot" tumors by reducing suppressive adenosine levels, maintaining immune-stimulatory extracellular ATP, and promoting effector T cell function and cytotoxicity against cancer cells. Preclinical studies have shown significant anti-tumor activity in models resistant to PD-1 antibodies. Developed for advanced solid tumors, ES014 has received IND clearance from both the FDA (United States) and CDE (China), with ongoing clinical trials in Phase 1/2 for various solid malignancies[1][3][4][5][6].
