ES015 is a first-in-class humanized monoclonal antibody designed to target Dickkopf-1 (DKK1), a secreted protein that plays a significant role in oncogenesis and immune evasion. DKK1 is a potent antagonist of the canonical Wnt/β-catenin signaling pathway; however, in the tumor microenvironment, its overexpression is associated with the inhibition of immune cell function, particularly natural killer (NK) cells and T cells, thereby facilitating tumor growth and progression. By specifically binding to and neutralizing DKK1, ES015 restores Wnt signaling in immune cells, enhancing their anti-tumor activity and potentially overcoming resistance to existing checkpoint inhibitors. Developed by Elpiscience Biopharmaceuticals, ES015 is currently undergoing Phase 1 clinical evaluation for patients with advanced solid tumors that exhibit high levels of DKK1 expression.
