ES015-2 is a bifunctional fusion protein designed to simultaneously target the programmed death-ligand 1 (PD-L1) and transforming growth factor-beta (TGF-β) pathways. Developed by Elpiscience, the molecule consists of a monoclonal antibody against PD-L1 fused to the extracellular domain of the TGF-β receptor II (TGF-βRII), which acts as a trap for TGF-β. By inhibiting PD-L1, ES015-2 aims to restore T-cell activity against tumor cells, while the TGF-β trap component neutralizes TGF-β in the tumor microenvironment, thereby reducing immunosuppression, fibrosis, and epithelial-mesenchymal transition (EMT). This dual mechanism is intended to overcome resistance to anti-PD-(L)1 monotherapies, particularly in cold tumors where TGF-β signaling is a dominant immune evasion strategy.
