ES019 is a bispecific antibody designed using Elpiscience's proprietary Bispecific Macrophage Engager (BiME) platform, targeting both Programmed death-ligand 1 (PD-L1) and Signal-regulatory protein alpha (SIRPα). The drug is intended to reactivate macrophages and T cells in the tumor microenvironment, thereby enhancing immune-mediated killing of cancer cells. By simultaneously blocking PD-L1—which enables tumor immune evasion—and SIRPα—which interacts with CD47 "don't eat me" signaling—ES019 aims to promote antigen-specific T cell activation and macrophage-mediated phagocytosis, overcoming resistance to current immune checkpoint inhibitors. ES019 is primarily being developed for the treatment of neoplasms, notably solid tumors. The originator and primary developer is Elpiscience Biopharma, with Astellas Pharma as a research, development, and potential manufacturing/commercialization partner. As of 2025, ES019 is at the preclinical/discovery stage, with no approved clinical indications.[1][3][5][6][7][9]
