ES102 is a first-in-class hexavalent OX40 agonist antibody developed by Elpiscience Biopharma (licensed from Inhibrx). It is designed to treat advanced solid tumors, including non-small cell lung cancer (NSCLC) and head and neck squamous cell carcinoma (HNSCC). Unlike traditional bivalent antibodies, ES102's hexavalent structure, composed of six OX40-binding single-domain antibodies, enables efficient clustering and activation of the OX40 (CD134) co-stimulatory receptor on T cells without the need for exogenous Fcγ receptor-mediated cross-linking. This mechanism enhances anti-tumor T-cell responses and has shown synergy with PD-1/PD-L1 inhibitors in preclinical models. The drug is currently being evaluated in clinical trials both as a monotherapy and in combination with checkpoint inhibitors.
