Overview

ES502 is a bispecific soluble T-cell receptor (TCR) fusion protein designed for the treatment of advanced solid tumors, specifically those harboring the RAS G12V mutation. Developed by Ruijin Hospital in collaboration with Essight Biopharma, the molecule consists of a soluble HLA-DP restricted TCR specific for the RAS G12V neoantigen, fused to an anti-CD3 antibody fragment and an Fc domain. This configuration allows the drug to act as a T-cell engager, redirecting T-cells to attack cancer cells presenting the RAS G12V peptide in the context of specific HLA-DP alleles. It is currently being evaluated in Phase 1 clinical trials for patients with advanced pancreatic cancer and other RAS G12V-positive solid tumors.
