Escherichia coli endotoxin, also known as lipopolysaccharide (LPS), is a major structural component of the outer membrane of the Gram-negative bacterium E. coli. [7, 8] It is a potent activator of the innate immune system. [1] The biological activity is primarily mediated by its lipid A component, which acts as a powerful agonist for the Toll-like receptor 4 (TLR4) in complex with myeloid differentiation factor 2 (MD-2). [3, 4, 10] This interaction triggers a robust pro-inflammatory cascade, leading to the release of cytokines like TNF-α and IL-6. [11] In clinical and biomedical research, highly purified E. coli endotoxin is used as a challenge agent in what is known as the human endotoxemia model. [1, 5, 15] It is administered to healthy volunteers in controlled, low doses (intravenously or via inhalation) to induce a transient, standardized systemic inflammatory response that mimics the early stages of sepsis. [1, 5] This allows researchers to investigate the pathophysiology of inflammation and to test the efficacy of novel anti-inflammatory therapeutic agents in a controlled human setting. [1, 15] It is considered a research tool rather than a therapeutic drug.
