Drug intelligence / Profile preview
eseba-vec + pembrolizumab (formerly HB-200 + pembrolizumab) is an investigational combination immunotherapy consisting of eseba-vec (HB-200) and the anti-PD-1 monoclonal antibody pembrolizumab. Eseba-vec is an arenavirus-based immunotherapy platform comprising two replicating, attenuated arenavirus vectors: HB-201 (derived from lymphocytic choriomeningitis virus) and HB-202 (derived from Pichinde virus). These vectors are engineered to express a non-oncogenic human papillomavirus type 16 (HPV16) E7/E6 fusion protein, designed to induce robust tumor-specific CD8+ T-cell responses against HPV16-positive tumors. Pembrolizumab acts as a checkpoint inhibitor to block the PD-1/PD-L1 pathway, thereby preventing T-cell inhibition and enhancing the vaccine-induced anti-tumor immune response. The combination is being evaluated in clinical trials for the treatment of patients with HPV16-positive recurrent or metastatic head and neck squamous cell carcinoma (HNSCC), particularly oropharyngeal squamous cell carcinoma.
