ESG401

ESG401 is an antibody-drug conjugate (ADC) composed of a humanized IgG1 monoclonal antibody targeting trophoblast cell-surface antigen 2 (TROP2), conjugated to the topoisomerase I inhibitor SN-38 via a proprietary stable-cleavable linker with a drug-to-antibody ratio (DAR) of 8. It is designed for the treatment of locally advanced or metastatic solid tumors, with particular focus on breast cancer subtypes such as triple-negative breast cancer (TNBC) and hormone receptor-positive/HER2-negative breast cancer. ESG401 has demonstrated promising antitumor activity and a favorable safety profile in heavily pretreated patients, including those with brain metastases. The most common adverse events are hematologic toxicities such as neutropenia and leukopenia, along with fatigue, nausea, and vomiting[1][2][5][7].
