Overview

**ESK1** is a human T-cell receptor (TCR)-mimic monoclonal antibody engineered to target the intracellular Wilms Tumor 1 (WT1) oncoprotein. It binds with high affinity (sub-nanomolar) to the RMF peptide (RMFPNAPYL) derived from WT1 presented in complex with HLA-A*02:01 and several other HLA-A*02 subtypes, enabling broader patient compatibility beyond typical TCR restrictions. ESK1 mediates antibody-dependent cell cytotoxicity (ADCC) against WT1-expressing cancer cells and has demonstrated therapeutic efficacy in mouse models of WT1-positive human cancers, such as leukemia and solid tumors. Originally developed by Eureka Therapeutics in collaboration with Memorial Sloan Kettering Cancer Center using a TCR-mimic platform, it was later advanced by Novartis as an Fc-enhanced version for intracellular WT1 targeting.[1][6]
