Overview

**ESO157-170** is a synthetic peptide vaccine corresponding to amino acids 157–170 of the NY-ESO-1 cancer-testis antigen. It is designed to induce cellular immune responses against NY-ESO-1-expressing tumors by stimulating both **CD4+ (helper)** and **CD8+ (cytotoxic) T cells**. Mechanistically, ESO157-170 acts as an epitope recognized by HLA-DP4-restricted CD4+ T cells and HLA-A2-/A24-restricted CD8+ T cells, promoting a Th1 biased cytokine response characterized by secretion of IFN-γ, TNF-α, and IL-2. ESO157-170 has been investigated in phase I clinical trials in patients with **epithelial ovarian cancer (EOC)**, formulated with Montanide ISA-51 adjuvant. Induced T cell clones were shown to recognize and destroy NY-ESO-1 expressing tumor cells. The vaccine's immunogenicity results in long-lived functional memory T cells, and it has potential utility in cancer immunotherapy, especially for tumors expressing NY-ESO-1[2][3][4].
