EAC-T is a combination pharmacological regimen investigated for the eradication of *Helicobacter pylori* and the prevention of gastrointestinal mucosal injury. The regimen consists of a standard triple therapy—comprising the proton pump inhibitor **esomeprazole** (E), and the antibiotics **amoxicillin** (A) and **clarithromycin** (C)—supplemented with **teprenone** (T), also known as geranylgeranylacetone. Esomeprazole reduces gastric acid secretion by inhibiting the H+/K+-ATPase pump, creating an environment conducive to antibiotic activity and mucosal healing. Amoxicillin and clarithromycin work synergistically to eliminate *H. pylori* infection. Teprenone acts as a cytoprotective agent by inducing heat shock proteins (specifically HSP70) and increasing the production of gastric mucus, thereby protecting the gastrointestinal lining from damage induced by the infection or by non-steroidal anti-inflammatory drugs (NSAIDs). This combination is particularly studied in patients with rheumatic diseases who require long-term NSAID therapy.
