Drug intelligence / Profile preview
Esomeprazole + buffered acetylsalicylic acid is a fixed-dose combination drug designed for oral administration that consists of **esomeprazole**, a proton pump inhibitor (PPI), and **acetylsalicylic acid** (ASA, commonly known as aspirin) in a buffered formulation. Esomeprazole acts by inhibiting the gastric proton pump (H+/K+ ATPase), reducing gastric acid secretion and protecting the gastric mucosa. Acetylsalicylic acid inhibits cyclooxygenase-1 (COX-1) in platelets, preventing platelet aggregation and reducing the risk of thrombosis, which is beneficial for cardiovascular protection. The buffered formulation of ASA is intended to reduce gastrointestinal irritation. This combination is used primarily for prevention of cardiovascular events in patients at increased risk of gastrointestinal complications from long-term aspirin therapy, such as peptic ulcers or bleeding[1][2][3][4]. Clinical trials have shown the combination does not impact the antiplatelet effects of aspirin, and its main benefit is a reduction in gastrointestinal damage associated with aspirin therapy[2][4]. The product has been studied in Phase 1 trials for peptic ulcer disease and cardiovascular protection[1][4]. Mechanism of action involves both **Gastric acid suppression** (esomeprazole) and **antiplatelet activity** (ASA). Side effects include increased bleeding tendency, indigestion, flatulence, nausea, and vomiting[3].
