ESP359 is a radioligand therapy (RLT) targeting Delta-like ligand 3 (DLL3), currently under development by Hefei China Resources Novartis Pharmaceutical, a joint venture between Novartis and China Resources. DLL3 is a Notch pathway ligand that is highly expressed on the surface of small cell lung cancer (SCLC) and other neuroendocrine tumors, while showing minimal expression in normal tissues, making it a promising target for precision oncology. As a radioligand therapy, ESP359 is designed to deliver a therapeutic radioisotope directly to DLL3-expressing malignant cells, where the resulting ionizing radiation induces DNA damage and cell death. The asset is currently in Phase 1 clinical development for the treatment of advanced solid tumors.
