espindolol

AGI-001 (espindolol) is an investigational oral small molecule and the S-enantiomer of the racemic beta-blocker pindolol. Developed by AGI Therapeutics, it was primarily evaluated for the treatment of functional dyspepsia (non-ulcer dyspepsia). Unlike the racemic mixture, which is used for cardiovascular conditions, espindolol acts as a potent antagonist at 5-HT1A and 5-HT1B serotonin receptors. This mechanism is hypothesized to improve gastric accommodation and reduce visceral hypersensitivity, thereby addressing symptoms such as early satiety, abdominal distension, and discomfort. The drug reached Phase II clinical trials in a randomized, double-blind, placebo-controlled study.
