Overview

ESR-114 is a selective, potent small molecule inhibitor of retinoic acid-related orphan nuclear receptor gamma t (RORγt), also known as nuclear receptor subfamily 1 group F member 3. It is being developed as a topical gel formulation designed to target its pharmacological activity to the skin with minimal systemic absorption. The mechanism of action involves antagonism of RORγt, which reduces production of the inflammatory cytokine IL-17—a key driver in psoriasis and other autoimmune disorders. The drug is primarily being investigated for the treatment of plaque psoriasis, with clinical trials reaching Phase 1/2a status in patients with mild-to-moderate disease[1][2][3][4].
