Overview

This investigational cancer vaccine is an adenoviral-vector-based immunotherapy targeting activating mutations in the ESR1 (estrogen receptor alpha) gene, specifically the Y537S and D538G variants. These mutations frequently emerge in patients with ER+ breast cancer following prolonged endocrine therapy, such as aromatase inhibitors, leading to constitutive, ligand-independent receptor activity and subsequent treatment resistance. Developed by researchers at Duke University, including Herbert K. Lyerly and Zachary C. Hartman, and licensed to Replicate Biosciences, the vaccine encodes mutant ESR1 proteins to elicit a neoantigen-specific T-cell mediated immune response. By training the immune system to recognize and eliminate cells expressing these specific resistance-driving mutations, the therapy aims to overcome endocrine resistance in metastatic ER+ breast cancer.
