Overview

Esterified butyrate, specifically delivered as butyrylated high-amylose maize starch (HAMSB), is a dietary intervention developed by the CSIRO Preventative Health National Research Flagship for the chemoprevention of colorectal cancer, particularly in patients with familial adenomatous polyposis (FAP). In this formulation, butyrate, a short-chain fatty acid (SCFA), is chemically esterified to a high-amylose maize starch backbone. This delivery system allows the butyrate to bypass the upper gastrointestinal tract and be released in the colon through bacterial fermentation of the starch. Once released, butyrate serves as a primary energy source for colonocytes and acts as a histone deacetylase (HDAC) inhibitor, promoting cell cycle arrest and apoptosis in neoplastic cells while supporting healthy colonic epithelial function.
