Overview

TNX-4300 (estianeptine) is an oral small molecule therapeutic being developed by Tonix Pharmaceuticals for the treatment of major depressive disorder (MDD), bipolar disorder, Alzheimer's disease, and Parkinson's disease. It is the (S)-enantiomer of tianeptine and acts as a dual agonist of peroxisome proliferator-activated receptors (PPAR) β/δ and γ. Unlike racemic tianeptine, TNX-4300 is designed to be free of µ-opioid receptor activity, which is associated with the (R)-enantiomer. The drug's mechanism of action involves restoring neuroplasticity and neurogenesis in neuronal tissue, representing a first-in-class approach to repairing atrophied neuronal connections in psychiatric and neurodegenerative conditions.
