Estradiol phenylpropionate is a synthetic estrogen ester, specifically the 17β phenylpropionate ester of estradiol, designed to provide prolonged estrogenic activity due to slow release from the intramuscular injection site.[3][1][2] It binds to **estrogen receptors** and acts as an agonist, modulating gene transcription and mediating multiple physiological effects typical of estrogen. Estradiol phenylpropionate has been used primarily for hormone replacement therapy (HRT) for menopausal symptoms, prevention of postmenopausal osteoporosis, treatment of estrogen deficiency (e.g., hypogonadism), and as part of formulations for transgender hormone therapy.[3][2][5] Its relatively slow release and long duration make it useful in injectable combination products for sustained hormone therapy. Common side effects include breast tenderness, nausea, headache, and fluid retention.[2] It is contraindicated in patients with estrogen-dependent neoplasia, breast cancer, severe liver disease, or during pregnancy.[2] Estradiol phenylpropionate is no longer widely marketed as a standalone agent, but has been present in combination formulations such as **Dimenformon Prolongatum**, **Mixogen**, and **Lynandron Prolongatum**.[3][5]
