Estrogen is a steroid hormone that was evaluated by the US Department of Veterans Affairs in a Phase 2 clinical trial (NCT00018343) for its potential to improve cognitive function in postmenopausal women with Alzheimer's disease. The therapy aims to leverage the neuroprotective properties of estrogen, which include the modulation of amyloid-beta processing, enhancement of synaptic plasticity, and improvement of cerebral blood flow. Estrogen acts as an agonist at estrogen receptors (ERα and ERβ) located throughout the brain, particularly in areas associated with memory and executive function. This repurposing effort was based on the hypothesis that estrogen deficiency following menopause contributes to cognitive decline and the pathology of Alzheimer's disease.
