Overview

ET019003-T is an autologous chimeric antigen receptor (CAR) T-cell therapy developed using the ARTEMIS platform, specifically designed to target the CD19 antigen expressed on B-cell malignancies. Unlike conventional CAR-T therapies that utilize single-chain variable fragments (scFv), ET019003-T employs a human anti-CD19 antibody Fab domain fused to the transmembrane and intracellular domains of a gamma-delta T-cell receptor (γδTCR). This design is intended to mimic natural T-cell activation and minimize the risk of tonic signaling and cytokine release syndrome. The cells are further modified with a Chimeric Signaling Receptor (CSR), known as ET190L1-CSR, expressed in trans. This CSR also binds to CD19 to activate a novel costimulatory domain, which enhances the proliferation and long-term persistence of the engineered T-cells in the patient. ET019003-T is primarily being evaluated for the treatment of relapsed or refractory CD19+ B-cell leukemia and lymphoma.
