ET6-21 is a novel, hydrophilic ligand designed to bind amyloid fibrils and amyloid deposits. It serves as a lead candidate for the preparation of amyloid-targeted liposomal nanoparticles. These nanoparticles, incorporating Gd chelates and Indocyanine green, are developed for the imaging of amyloid plaque via magnetic resonance imaging (MRI) and near-infrared microscopy. In preclinical studies using Tg2576 and TetO/APP mouse models of amyloid deposition, ET6-21-containing nanoparticles successfully crossed the blood-brain barrier and bound to amyloid plaques, enabling elevated MRI signals in amyloid-positive mice. This suggests its potential as a diagnostic agent for conditions involving amyloid accumulation, such as Alzheimer's disease.
