Drug intelligence / Profile preview
Etaftoron (also known as etaftorone or OSI-1211) is a lipid-soluble 1,4-dihydropyridine (DHP) derivative developed by the Latvian Institute of Organic Synthesis. Unlike traditional dihydropyridines, etaftoron exhibits very weak calcium channel blocking activity. Instead, it acts primarily as a mitochondrial modulator, peroxynitrite scavenger, and DNA-binding agent. In preclinical models of streptozotocin-induced diabetes mellitus, etaftoron has been shown to reduce pathological nitric oxide (NO) hyperproduction, downregulate inducible nitric oxide synthase (iNOS) and xanthine oxidoreductase (XOR) expression, and upregulate endothelial nitric oxide synthase (eNOS) expression, thereby demonstrating potential protective effects against diabetic nephropathy and oxidative stress-induced DNA damage.
