Etanercept and adalimumab are both biologic disease-modifying antirheumatic drugs that act as tumor necrosis factor-alpha inhibitors. They are used to treat a range of autoimmune and inflammatory diseases, including rheumatoid arthritis, ankylosing spondylitis, psoriatic arthritis, juvenile idiopathic arthritis, plaque psoriasis, Crohn’s disease, ulcerative colitis, hidradenitis suppurativa, and uveitis. Etanercept is a dimeric fusion protein consisting of the extracellular ligand-binding portion of the human p75 TNF receptor linked to the Fc portion of human IgG1. Adalimumab is a fully human monoclonal antibody targeting TNF-alpha. Both drugs block the activity of TNF-alpha—a cytokine involved in systemic inflammation—thereby reducing inflammation and halting disease progression[6][8][10]. Clinical studies show comparable efficacy and safety between these two agents for their approved indications[1][2][4].
