ETBX-051 is a therapeutic cancer vaccine composed of a replication-defective, serotype 5 adenovirus (Ad5) vector engineered to express the human tumor-associated antigen brachyury. The viral genes early 1 (E1), early 2b (E2b), and early 3 (E3) are deleted to prevent replication. The primary mechanism is immunostimulation, aiming to induce cytotoxic T-cell responses against tumor cells expressing the transcription factor brachyury, which is implicated in epithelial-mesenchymal transition and metastasis in various cancers. ETBX-051 was originally developed by Etubics Corporation and further developed by ImmunityBio and the National Cancer Institute for use as monotherapy or as part of a multi-antigen regimen with other adenoviral vaccines targeting CEA (ETBX-011) and MUC1 (ETBX-061). It has been investigated primarily for solid tumors including prostate cancer, colorectal cancer, pancreatic cancer, triple-negative breast cancer, non-small cell lung cancer, and metastatic cancers[1][3][4][6].
