ETBX-061 is a next-generation cancer vaccine based on a replication-defective adenovirus serotype 5 (Ad5) vector, engineered with deletions in the E1, E2b, and E3 genes. It encodes the human glycoprotein mucin 1 (MUC1), a tumor-associated antigen overexpressed in various cancers. The vaccine is designed to stimulate an immune response—specifically CD4+ and CD8+ T-cell responses—against cells expressing MUC1. ETBX-061 has been evaluated as part of a multitargeted regimen alongside other adenoviral vaccines targeting CEA and brachyury antigens for advanced cancers such as colorectal cancer, liver cancer, pancreatic cancer, and triple negative breast cancer. The regimen has demonstrated safety and immunogenicity in early-phase clinical trials[2][5][6][8].
