Overview

ETBX-061 + adenoviral MUC1 vaccine is a combination immunotherapeutic regimen consisting of the investigational agent ETBX-061, which is an adenovirus serotype 5 (Ad5)-based recombinant cancer vaccine encoding the tumor-associated antigen mucin 1 (MUC1). This combination is often administered alongside other similar Ad5-based vaccines targeting additional antigens such as CEA and brachyury in clinical trials for advanced cancers. The mechanism of action involves stimulating the immune system to generate cytotoxic T-cell responses specifically against cells expressing MUC1, a glycoprotein overexpressed and aberrantly glycosylated in many epithelial cancers. The goal is to induce robust anti-tumor immunity by presenting the MUC1 antigen via a viral vector platform that enhances immunogenicity[2][4][5]. Clinical studies have demonstrated that this approach can safely elicit antigen-specific CD4+ and CD8+ T-cell responses in patients with advanced malignancies[4][5].
