Overview

ETBX-071 (also known as Ad5-Brachyury) is a therapeutic cancer vaccine developed by ImmunityBio in collaboration with the National Cancer Institute (NCI). It utilizes a second-generation adenovirus type 5 (Ad5) platform that features deletions in the E1, E2b, and E3 regions. These modifications are designed to minimize the immune response against the viral vector itself, thereby reducing neutralization by pre-existing Ad5 antibodies and allowing for repeated booster administrations. The vaccine delivers a transgene encoding Brachyury (T), a transcription factor that is a master regulator of epithelial-mesenchymal transition (EMT). Brachyury is highly expressed in various metastatic and treatment-resistant solid tumors—including prostate cancer, chordoma, and colorectal cancer—but has minimal expression in normal adult tissues. By expressing this antigen, ETBX-071 aims to stimulate a robust, tumor-specific CD8+ T-cell response. It is frequently studied in combination with other immunotherapies, such as the IL-15 superagonist N-803, to enhance its clinical efficacy.
