Overview

ETC-101 is a bispecific T-cell engager (TCR-bispecific) immunotherapy designed to target the PReferentially expressed Antigen in MElanoma (PRAME). Developed by Etcembly using its proprietary generative AI platform, EMLy™, ETC-101 is engineered to overcome the limitations of traditional TCR discovery by optimizing affinity and specificity for the PRAME-peptide-MHC complex. PRAME is a cancer-testis antigen that is highly expressed in a wide range of solid tumors—including melanoma, non-small cell lung cancer (NSCLC), and ovarian cancer—but has minimal expression in healthy adult tissues, making it an ideal therapeutic target. The molecule functions by simultaneously binding to PRAME on tumor cells and the CD3 receptor on T-cells, thereby redirecting the immune system to selectively recognize and eliminate malignant cells. It is recognized as one of the first immunotherapy candidates designed primarily through generative AI technology.
