Etevritamab (AMG 596) is a bispecific T-cell engager (BiTE) antibody developed by Amgen for the treatment of glioblastoma and other malignant gliomas. It is engineered to specifically target the mutant epidermal growth factor receptor variant III (EGFRvIII), a tumor-specific antigen that is not expressed in normal tissues, thereby minimizing off-target toxicity. The molecule simultaneously binds to EGFRvIII on the surface of tumor cells and the CD3 epsilon subunit of the T-cell receptor complex on T cells. This dual binding brings T cells into close proximity with tumor cells, triggering T-cell activation, cytokine release, and the subsequent cytotoxic destruction of the EGFRvIII-expressing cancer cells. Clinical development of etevritamab reached Phase 1/1b, where it was studied as a monotherapy and in combination with the PD-1 inhibitor AMG 404, before being discontinued from Amgen's pipeline.
