ETH47 is an investigational mRNA-based therapeutic developed by Ethris for the treatment of respiratory diseases, primarily uncontrolled asthma. It encodes type III interferon (interferon lambda, IFNλ), a protein essential for antiviral immunity in the respiratory tract. Delivered via nasal spray or inhalation using Ethris’ proprietary SNIM RNA and SNaP LNP platforms, ETH47 induces local production of IFNλ at mucosal surfaces to enhance innate immune defense and inhibit viral replication at virus entry sites. Phase 1 clinical trials demonstrated that intranasal administration of ETH47 is generally safe and well-tolerated with no severe adverse events or systemic bioavailability. The drug showed dose-dependent target engagement, increasing IFNλ levels in the nasal lining fluid and activating downstream antiviral interferon-stimulated genes (ISGs). Its mechanism aims to address the upstream triggers of asthma exacerbations—primarily viral infections such as rhinovirus—and it is being further developed for broader applications in respiratory viral infections including influenza and COVID-19[1][2][3][4][5][7][8].
