Overview

Ethenzamide is a small molecule non-opioid analgesic and anti-inflammatory drug used for the relief of pain (including headache and toothache), fever, and other minor aches. It is commonly available as an over-the-counter medication in East Asia—especially Japan—where it is often found in combination with caffeine and acetaminophen for indications such as headache, menstrual cramps, toothache, and fever[1][3][7]. Ethenzamide acts primarily via antagonism of the 5-hydroxytryptamine (serotonin) 2B receptor (5HT2B), with additional modest inhibitory effects on monoamine oxidase-A and transient receptor potential vanilloid 1 channel; it does not inhibit cyclooxygenase-1 or -2[6][8]. In vivo it is metabolized to salicylamide[3]. It has been used both alone and in combination with other NSAIDs for symptomatic relief of cold symptoms[5].
