Overview
Ethinyl estradiol + chlormadinone acetate is a combined oral contraceptive containing two active ingredients: ethinyl estradiol (a synthetic estrogen) and chlormadinone acetate (a progestin with antiandrogenic properties)[1][2][8]. This combination is used primarily for contraception and is approved in Europe but not in the United States[8]. The mechanism of action involves suppression of ovulation by inhibiting gonadotropin secretion; ethinyl estradiol acts as an agonist at the estrogen receptor to suppress follicle-stimulating hormone (FSH) and luteinizing hormone (LH), while chlormadinone acetate acts as an agonist at the progesterone receptor and also blocks androgen receptors[4][5]. Together they prevent ovulation, thicken cervical mucus to inhibit sperm penetration, alter endometrial lining to prevent implantation, and provide antiandrogenic effects beneficial for conditions like acne[2][5][8]. The combination has been shown effective for contraception and has demonstrated efficacy in treating mild-to-moderate papulopustular acne[2].
