ETHY-001 is a half-life extended monoclonal antibody antagonist of the thyroid-stimulating hormone receptor (TSHR) currently in development for the treatment of Graves' hyperthyroidism and thyroid eye disease (TED). The antibody is designed to directly block pathogenic stimulating autoantibodies (TSAbs) that inappropriately hyperstimulate TSHR in thyroid and orbital tissues. In vitro studies have demonstrated that ETHY-001 binds to TSHR with high affinity and specificity, fully blocking TSHR activation induced by patient-derived polyclonal TSAbs and the monoclonal antibody M22. Furthermore, it has shown the ability to completely inhibit M22-induced secretion of hyaluronan and IL-6 from TED patient-derived orbital fibroblasts, outperforming IGF-1R antagonism in certain assays. By targeting the causal disease process of Graves' disease, ETHY-001 aims to provide a comprehensive treatment option for both the hyperthyroid and ophthalmic manifestations of the disorder.
