Overview

Etigilimab is a humanized IgG1 monoclonal antibody that targets T-cell immunoreceptor with Ig and ITIM domains (TIGIT), an immune checkpoint receptor expressed on T cells and regulatory T cells. By binding to TIGIT, etigilimab blocks its interaction with the poliovirus receptor (PVR), thereby inhibiting downstream signaling that suppresses immune responses. This blockade leads to reduced regulatory T cell (Treg) frequency and increased CD8/Treg ratio in peripheral blood, promoting enhanced proliferation and cytokine function of effector T cells. Etigilimab is being developed primarily for the treatment of advanced or metastatic solid tumors—including ovarian cancer, fallopian tube cancer, peritoneal cancer—and has been studied in combination with nivolumab in clinical trials. The drug was originally developed by OncoMed Pharmaceuticals and is currently under development by Mereo BioPharma[1][2][3][6].
