Overview

Etoposide + thiotepa is a combination chemotherapy regimen consisting of two small molecule cytotoxic agents, etoposide and thiotepa. Etoposide is a topoisomerase II inhibitor that induces DNA strand breaks, leading to apoptosis in rapidly dividing cells. Thiotepa is an alkylating agent that crosslinks DNA strands, inhibiting DNA replication and transcription. This combination has been used primarily as high-dose chemotherapy (HDC) for conditioning prior to autologous stem cell or bone marrow transplantation in patients with high-risk or recurrent primary brain tumors, relapsed/refractory germ cell tumors, and certain lymphomas. The regimen aims to maximize tumor cytoreduction before hematopoietic rescue[1][2][7].
