Etoposide and Paclitaxel

Etoposide and paclitaxel are two chemotherapy agents with different mechanisms of action that are used in combination for treating various cancers. Etoposide is a podophyllotoxin derivative that inhibits DNA synthesis by forming a complex with topoisomerase II and DNA, inducing breaks in double-stranded DNA and preventing repair. Paclitaxel is a taxane that stabilizes microtubules, preventing their breakdown during cell division, which leads to cell death. The combination has been studied in various cancer types, particularly small cell lung cancer and testicular cancer. The sequence of administration appears to be important, as different sequences can affect both efficacy and toxicity profiles.
